Introduction:

동물 조직(animal tissues) 및 배양 세포(cultured cells)에서 단백질을 추출하고 정제하는 과정에서 방출될 수 있는 내인성 프로테아제(endogenous proteases)에 의한 단백질 분해를 효과적으로 방지하여, 단백질을 완벽하게 보호하도록 설계되었습니다.

단백질 추출 과정에서 조직 또는 세포를 용해시키는 lysis buffer에는 리소좀(lysosome)에서 유래한 다양한 효소가 존재하게 되며, 비활성 상태의 프로테아제들 또한 활성화됩니다. 활성화된 프로테아제는 주변 단백질을 분해하기 시작하므로, 단백질 손실을 방지하기 위해서는 반드시 Protease Inhibitor를 lysis buffer에 첨가해야 합니다.

동물(포유류), 식물, 곰팡이(mould), 박테리아(bacterial) 등 다양한 생물에서 유래하는 프로테아제의 구성은 서로 다르며, 생물의 성장 단계와 생리적 상태에 따라서도 변화할 수 있습니다. 따라서 다양한 종류의 단백질을 추출하고 전기영동(Electrophoresis) 분석을 수행할 때는 목적에 적합한 Protease Inhibitor Cocktail을 선택하는 것이 매우 중요합니다.
